ARA-290 is a synthetic 11-amino acid peptide derived from the helix-B surface domain of Erythropoietin (EPO). Mechanism of Action: The Innate Repair Receptor (IRR)

ARA-290 functions through a highly specific receptor interaction that triggers an immediate anti-inflammatory cascade.

  1. Heteromeric Binding: The peptide acts as a specific agonist for the Innate Repair Receptor (IRR), a heterocomplex formed by the Erythropoietin Receptor (EPOR) and the Beta-Common Receptor (CD131 or βc).

  2. Signal Transduction: Upon binding to the IRR, ARA-290 initiates downstream signaling that inhibits the activation of NF-κB (Nuclear Factor kappa-light-chain-enhancer of activated B cells).

  3. Pathological Interruption: By blocking NF-κB, the peptide effectively "turns off" the production of pro-inflammatory cytokines (TNF-α, IL-6) and blocks the pain signals associated with neuropathic inflammation. Crucially, because it does not bind to the EPOR homodimer, it has zero effect on hemoglobin levels.
